Sure, as long as it is a dry area. Kitchen backsplash for example.

I would rough up the drywall a bit with some sandpaper to make the surface better for the thinset to bind to.

Depends on the type of paint: if it's semi or high gloss then I would defintately rough up the surface prior to laying tile.

I would also make sure the surface is flat no bumps or paint drips; if you find any just lightly sand them.
